Assessment of the metabolism of different strains of Bacillus megaterium BABT
Suarez,Carlos Alberto Galeano; Montano,Inti Doraci Cavalcanti; Nucci,Edson Romano; Iemma,Monica Rosas da Costa; Giordano,Raquel de Lima Camargo; Giordano,Roberto de Campos.
Bacillus megaterium is a promising host for expression of heterologous proteins. This paper reports the nutrient consumption patterns and production of metabolites for three different strains of B. megaterium, ATCC 14945, QMB 1551 and PV 361, which is QMB 1551 with seven constitutive plasmids deleted. 14 h cultivations in agitated flasks were run, for two different media: A (LB plus 10g/L glucose) and B (medium A, with the yeast extract replaced by tryptone). Strains PV361 and QMB 1551 showed higher maximum specific growth rates in medium B, reaching 0.42 h-1 and 0.48 h-1 respectively. The main by-products of the glucose overflow mechanism were acetate and lactate, for all three strains, which had preferential amino acids for substrate: Ala, Asp, Glu, Ser....

Early Changes in Soil Metabolic Diversity and Bacterial Community Structure in Sugarcane under Two Harvest Management Systems Rev. Bras. Ciênc. Solo
Azevedo,Lucas Carvalho Basilio; Morais,Marcio; Lambais,Marcio Rodrigues.
Preharvest burning is widely used in Brazil for sugarcane cropping. However, due to environmental restrictions, harvest without burning is becoming the predominant option. Consequently, changes in the microbial community are expected from crop residue accumulation on the soil surface, as well as alterations in soil metabolic diversity as of the first harvest. Because biological properties respond quickly and can be used to monitor environmental changes, we evaluated soil metabolic diversity and bacterial community structure after the first harvest under sugarcane management without burning compared to management with preharvest burning. Soil samples were collected under three sugarcane varieties (SP813250, SP801842 and RB72454) and two harvest management...

Prado,R.O.F.; Morales,B.J.E.; García,M.L.J.; Molina-Ochoa,J.; Valpuesta,S.G.; Hernández,R.J.A.; García,C.A.C..
ABSTRACT English Thoroughbred horses, are widespread in Mexico and due to the lack of data on their exercise physiology, it is important to conduct exercise tests in order to obtain information the effects of exercise on more essential cations/anions in blood serum, as these horses are submitted to constant efforts. The study was carried out with 150 blood samples of English Thoroughbred horses clinically healthy. The blood sample collection was performed during three periods: 1) rest, 2) 30min after exercise (speed race of 12km/h for 30min with no rest) and 3) 60min after exercise. Mean values were calculated for cations (sodium and potassium) and anions (chloride and bicarbonate). The resulting data set was analyzed using Gaussian distribution and...

Extraction and Simultaneous Determination of Glyphosate, AMPA and Compounds of the Shikimic Acid Pathway in Plants Planta Daninha
Gomes,G.L.G.C.; Carbonari,C.A.; Velini,E.D.; Trindade,M.L.B.; Silva,J.R.M..
This study has aimed to develop a method for simultaneous extraction and determination by liquid chromatography and mass spectrometry (LC-MS/MS) of glyphosate, aminomethylphosphonic acid (AMPA), shikimic acid, quinic acid, phenylalanine, tyrosine and tryptophan. For the joint analysis of these compounds the best conditions of ionization in mass spectrometry and for chromatographic separation of the compounds were selected. Calibration curves and linearity ranges were also determined for each compound. Different extraction systems of the compounds were tested from plant tissues collected from sugarcane (Saccharum officinarum) and eucalyptus (Eucalyptus urophylla platiphylla) plants two days after the glyphosate application at the dose of 720 g a.e. ha-1....

Farming and soil urban occupation in the water quality of Jaboticabal and Cerradinho streams Ciência e Agrotecnologia
Silveira,Géssica Aparecida; Saran,Luciana Maria; Melo,Wanderley José de; Alves,Lucia Maria Carareto.
ABSTRACT Since the end of the twentieth century, 100% of the urban sewage from the city of Jaboticabal has been collected by interceptors and routed to a treatment plant. Between 1999 and 2000, studies on the effect of this environmental care showed that it did not efficiently clean up the water from the two streams that run through the city and flow into an agricultural area. This paper focuses on assessing the influence of soil use on the water quality of surface waters from these two streams. The study was conducted 15 years after the implementation of sewage interceptors. The sampling dates were bimonthly at eight points (P1 to P8) in the Cerradinho and Jaboticabal streams, in Jaboticabal (São Paulo State, Brazil). P1 was located at the source of the...

Feeding dairy cows with soybean by-products: effects on metabolic profile Ciência Rural
Alves,Marilia; González,Félix; Carvalho,Nelcy; Mühlbach,Paulo; Lima,Verônica; Conceição,Tiago Reis; Wald,Vera.
Soybean by-products are currently used in Rio Grande do Sul, Brazil, as a source of protein in dairy cows. However, the high protein breakdown of this feed source in the rumen causes loss of the intrinsic quality of the protein and increases plasma urea with deleterious consequences to animal metabolism. Thermal treatment of soybean can overcome this limitation. The aim of this study was to evaluate the metabolism of dairy cows fed with thermally treated soybean and raw soybean, through the analyses of the metabolic profile of plasma and milk. Twelve Holstein cows in mid-lactation period were studied using four treatments: commercial concentrated of protein, soybean meal, raw soybean and roasted soybean. Protein and urea levels in milk were measured in...

Metabolic Profiles in Ovulatory and Anovulatory Primiparous Dairy Cows During the First Follicular Wave Postpartum OAK
Kawashima, Chiho; Sakaguchi, Minoru; Suzuki, Takahiro; Sasamoto, Yoshihiko; Takahashi, Yoshiyuki; Matsui, Motozumi; Miyamoto, Akio.
Metabolic hormones affect ovarian function in the cow. However, the relationship between metabolic factors and ovarian function is not clear in the postpartum primiparous cow because they are still growing. The aim of the present study was to investigate in detail the time-dependent profile of the metabolic hormones, metabolites, and milk yields of ovulatory and anovulatory primiparous cows during the first follicular wave postpartum. We used 16 primiparous Holstein cows and obtained blood samples for the profiles of metabolites (glucose; non-esterified fatty acid, NEFA; ketone body; total cholesterol; and aspartate aminotransferase), metabolic hormones (growth hormone, GH; insulin-like growth factor-I, IGF-1; and insulin), and progesterone every other day...
